AQP4 in cerebral ischemia

Project description:

Edema is a major complication in brain ischemia such as after stroke. We have found that AQP4 is dramatically downregulated in the core of an experimental stroke. This downregulation may hamper dissolution of the postischemic edema and thus exacerbate the neurological impairment after stroke. The mechanisms of AQP4 downregulation - and the consequences of this in regard to extracellular volume control – is not known. In collaboration with Dr Sykova we will monitor the changes in extracellular space volume in the reperfusion phase of an ischemic insult and correlate these changes to alterations in AQP4 expression. With Dr Dirnagl we will employ a proteomics strategy to resolve how ischemia affects the composition of the dystrophin-associated protein complex (DAP) to which AQP4 is normally anchored. The latter experiments will help determine why AQP4 expression is so sensitive to ischemia.
