Topic Tubby and tubby-like-proteins (TULPs) are members of a novel gene family with an hitherto unknown function. Splicing defects in the carboxy-terminal half of two family members have been described to cause retinal degeneration in mice (tub) and humans(TULP1). (1) Evaluation of TULP3 as a candidate gene for an early-onset retinal degeneration in dogs. We cloned the gene and are now trying to put it on a canine radiation hybrid panel to test whether it maps within the minimal region of this disease. Also, we will compare the cDNA sequences and expression levels of TULP3 in affected and unaffected dogs. (2) Identification of the TULP1 promotor region. By screening BAC and P1 libraries we are trying to characterize the promotor region of the TULP1 gene in the mouse. As this gene is almost exclusively expressed in photoreceptor cells, we are looking for active binding-sites of retinal-specific transcription factors. The rationale is to understand how the transcriptional regulation works in those cells and how this might contribute to the pathogenesis of retinal diseases. |

Reactions to the U.S. Experience I had been told that Maine is a rugged landscape which can be pretty cold and unpleasant ... but the opposite turned out to be true! With the beautiful Acadia National Park just in walking distance of our house and exceptional weather for that time of the year, hiking and biking was possible until late in November. The few people you meet on your way seem taciturn but really friendly. Maybe because they are a little bit like the natives from the Northfrisian islands, I felt home right from the beginning.At the Jackson Laboratory, I was supported in any possible way. It was demanding, but it was real fun. (Thanks to Patsy, Juergen, Aki and the rest of the crew!) |
